RATIONALE Aging is associated with widespread changes in genome-wide patterns of DNA methylation. The methylation levels at CpG site in the gene ELOVL2 are strongly correlated with chronological age have been previously identified. However, the majority of these studies have focused on populations of western. METHODS We evaluated the relationship between the site cg16867657, annotated to the gene ELOVL2 and chronological age in prospective COPD cohort. DNA methylation levels of ELOVL2 in blood were measured in 180 adults. Pyrosequencing was performed to measure DNA methylation of ELOV2. The association with DNA methylation and chronologic age was analyzed using logistic regression analysis with adjustment for extrinsic environmental factors. RESULTS The mean (SD) age of study patients was 73.1 (7.8) years, and 25% were female There was an age-dependent increase in the methylation levels at the site cg16867657 (ℬ=0.36,; 95% CI: 0.27-0.58). After adjusting with gender, BMI, smoking amount, intake of folate, calories, zinc, and cholesterol, the DNA methylation of ELOVL2 was significantly increased according to age (ℬ=0.34; 95% CI: 0.15-0.60). CONCLUSIONS We confirm that an age associated site in the gene ELOVL2 shows a remarkably congruent relationship with aging in middle and old aged Korean. These results, by validating previous relevant studies, seem to provide additional support for further consideration of cg16867657 methylation as a true biological marker for age across the full spectrum of human diversity.
