Pulmonary fibrosis is a prototype of chronic, progressive, and fibrotic lung disease. Some studies have revealed that fibrotic process is associated with epithelial-mesenchymal(EMT). Currently, biomaterials have been testified to have both hard and soft tissue repair function, and it is a hot issue in the area of treatment to pulmonary fibrosis. But it is still unclear how calcium silicate(CS) participate in the process of pulmonary regeneration. Therefore, we are planning to explore the effects of CS on pulmonary epithelial cells(A549) and EMT. In vitro, CS extract was diluted at the ratios of 1/2,1/4,1/8,1/16,1/32,1/64 and 1/128, results showed that the CS extract could stimulate the proliferations of pulmonary epithelial cells in the ratios of 1/8,1/16,1/32, we also find that it increases the apoptosis of A549 in the ratios of 1/2. We use TGF-beta to induce the EMT of A549. Western blotting tested the EMT showed that CS extract has neither positive nor negative effect on the A549. In this exploratory test we clarified the crucial role of CS in lung regeneration and guide a novel pathway to the treatment of pulmonary fibrosis.
