Objective: To investigate the mechanism of B cell activating factor receptor (BAFFR) in immune response of Pneumocystis carinii infection in mice. Methods: BAFFR-/- mice and WT mice model were infected with pneumocystis by intratracheal injection, RT-PCR was used to detect pneumocystis load of lung tissue at different time points (2W, 3W, 4W, 5W) , the quantity and proportion of mature B cell subsets and T cell subsets was analyzed by flow cytometry. Results: Pneumocystis load in lung tissue showed no significant difference at 2W,3W,4W,increased at 5W (BAFFR-/- vs WT, P=0.008), the quantity of CD19+CD3- B cells in lung tissue, peripheral blood and spleen of BAFFR-/- PCP mice were significantly decreased compared with WT PCP mice at different time points, there were significant difference of B cell subsets in peripheral blood and spleen, while no obvious difference of each subgroup in the lung tissue, the quantity of CD4+ T cells,CD8+ T cells,Th17 cells were significantly increased (BAFFR-/- PCP vs, WT PCP, P=0.0086, P=0.0045,P=0.0012) at 2W, the amount of CD4+ T cells, CD8+ T cell showed no significant difference at 3W,4W, 5W.Conclusion: The deletion of B cell activating factor receptor impeded the elimination of pneumocystis in mice, B lymphocytes played a role in the elimination of pneumocystis in mice.
